Searching for Suitable Conditions for Proliferation and Differentiation of Stem and Progenitor Cells Isolated from CNS of Human and Animal Brain

The technology for preparation of cultures of stem and progenitor cells (SPC] from neurogenic zones of human and animal brain was developed. Optimum medium composition for proliferation of neural SPC forming neurospheres in suspension culture was determined. Characterization of morphology and phenotype of cultured cells with use of monoclonal antibodies for markers of stem, progenitor and mature cells was performed. The ability of SPC to grow and accumulate biomass during three passages was established. Comparative evaluation of growth intensity of neurospheres isolated from different regions of human and animal brain was pursued. More intensive proliferation of SPC isolated from animal subependymal zone of brain ventricles and olfactory bulb was recognized.
