We have analyzed karyotype of early-passaged human mesenchymal stem cells (hMSC) compared to that of peripheral blood lymphocytes from the same individuals. Karyotyping was performed on cultures of hMSC and PHA-stimulated peripheral blood lymphocytes donated by 8 healthy men and women volunteers. Cytogenetic analysis of hMSC cultures after each passage, from fourth to seventh, revealed no metaphase plates with abnormal chromosome number and/or structure. In all cases we established normal karyotype, corresponding karyotype of PHA-stimulated lymphocytes from the same individuals. Thus, in early-passaged cultures of hMSC from donors with normal karyotype in lymphocytes, no cells with chromosomal and/or genomic mutations were detected.
